A completely new base material for low pressure, pipette-tip tryptic digestion of proteins has been developed by GL Sciences. This unique material consists of Trypsin (TPCK) immobilized on the surface of a porous silica monolithic disc, which is embedded in a pipette tip. Tips are available in 10-200uL sizes in packages of 8 or 96.![]() ![]() Figure 1 The chromatogram above shows an HPLC analysis of denatured transferrin before and after digestion with MonoTip Trypsin. The 0.1mg/ml solution of transferrin was cycled approximately 10 times through a MonoTip Trypsin pipette tip at room temperature. Figure 2 The chromatograms below compare the results of tryptic digest in under 15 minutes using MonoTip Trypsin at room temperature versus traditional bulk digestion for 15 hours at 37°C. The top chromatogram shows the results from the MonoTip digest and the lower chromatogram shows the results from the traditional bulk, in-solution digest. As indicated, the results show MonoTip Trypsin does a very efficient digestion in a fraction of the time of traditional methods. (click on image below to enlarge) ![]() | ||||
